We have re-engineered the molecular architecture of the polymerase chain reaction, empowering it to process the information from multiple biomarkers simultaneously. Rather than rely on laborious, repetitive protocols or expensive, specialized equipment, our innovative approach combines the power of gene expression signature diagnosis with the ease and familiarity of a single PCR reaction.

 

RNA diagnosis: a compelling modality slowed down by legacy technologies.

Transcriptional signatures, or gene expression signatures, offer a modern approach to diagnosis of complex diseases and conditions. Rather than relying on one or two biomarkers, these signatures rely on a pattern of differential expression exhibited by a panel of numerous mRNA or miRNA targets. A patient can be diagnosed based on whether this panel looks more like the “healthy” pattern or more like the “disease” pattern. These signatures offer a powerful window into patient physiology, but translating them to the clinic has been hampered by traditional approaches.

Legacy approaches for measuring gene expression signatures were built for the lab, not the clinic, and it shows. Running a dozen qPCR reactions in parallel? Limited throughput, poor reproducibility. Sequencing? Microarrays? High up-front and reagent costs, testing bottlenecks, specialized and laborious sample preparation, and sophisticated analysis needed for interpretation. The latest all-in-one device? Minimal flexibility, shallow market penetration, and questionable durability. The drawbacks of these technologies have hamstrung the clinical translation of this powerful diagnostic modality.

The science under the hood.

Legacy technologies rely on measuring each RNA sequence one-by-one and then combining this information later. But a clinician doesn’t care whether GBP6 is high, IFI44L is low, and TMCC1 is a little high, they just want to know if the patient has the disease! So why not couple measurement and interpretation into a single step?

The PCRchitectur platform combines the nuance of biology with the power of machine learning on the molecular level. We’ve hacked the machinery of PCR to interrogate each transcript in a sample, producing one fluorescent signal for every target that matches the “healthy” pattern and a second signal for every target that matches the “disease” pattern. The better the match, the stronger the signal. The result is a simple readout where the stronger of the two colors indicates the diagnosis.
